antibody/antigen protocols

The Center has been collaborating with Abgenix to develop protocols for charactering monoclonal antibodies. Recently we submitted a patent application on a new method for analyzing crude antibodies in a screening format. These methods make it possible to rank antigen binding characteristics based on association and dissociation rates from a single antigen concentration. Currently using Biacore technology, ~50 to 100 monoclonal antibody samples may be screened from crude supernatants within a 24-hour period.

The Center is also optimizing methods for determining binding constants for extremely high affinity (pM) antibodies. Several manuscripts are in preparation that highlight both of these methods.
