array data

To test the reliability of the array sensor, one protein was spotted 144 times in a 12x12 grid within a single flow cell. A binding partner was injected across the array at 5 different concentrations. Each data set was fit to a 1:1 interaction model.

Examples of binding responses at 108 of the spots are shown on the right. The average and standard deviation of the binding constants observed throughout the 144-sample grid were:

ka = (1.68 ± 0.2) e5 M-1s-1
kd = (1.57 ± 0.1) e-3 s-1
KD = 9.4 ± 1 nM

On average the binding constants varied by only 10% between spots.
