Allelic Imbalance (LOH) In the following normal/tumor pairs, the first pair shows no difference in relative allele peak height between normal and tumor. In the second pair, one allele is obviously reduced. In some cases, an allele is completely missing. In others, there is a mixed population of tumor and normal with allele loss ranging from obvious to subtle. These cases of allelic imbalance are handled well with fluorescent technology where the areas under the peaks can be directly determined and scored as in the following example:
Ratios lower than 0.5 or higher than 2.0 are strongly indicative of LOH, however the researcher needs to determine a statistical cutoff based on type of error tolerated. |
